The BM (Basic Medium) was developed for the germination of Western European orchids by Van Waes and Debergh (1986). After testing 23 different species, the authors concluded that European orchids are more sensitive to salts compared to tropical orchids. The germination rate was higher on less saline media. In BM1 or BM2, nitrogen was not added via inorganic sources, but via casein hydrolysate and glutamine.
BM2 is composed like BM1, but additionally contains 0.88 µM BAP. This cytokinin led to an increase in the germination rate in some species and a decrease in others. In the following list, the species with increased germination rates are assigned to the respective medium:
BM1:
Anacamptis pyramidalis
Dactylorhiza spp.
Gymnadenia spp.
Listera ovata Ophrys spp.
Orchis spp.
Platanthera spp.
Spiranthes spp.
BM2:
Aceras anthropophorum
Cephalanthera spp.
Cypripedium calceolus
Epipactis spp.
Limodorum abortivum
A purified casein hydrolysate with lowered salt and high nitrogen content is used for this medium. This is not identical to the hydrolysate used by Van Waes and Debergh. Please contact us if you require the formula with the variant originally used.
